Most likely, King Boo will return as the game's final boss. People also say that this game is more puzzle orientated. Found a new image of Luigi in an icy room with Toad maybe following him:

Okay, this is what has been confirmed so far:

Characters

    Luigi
    Professor E. Gadd
    Toad

Species

    Boo
    Ghosts

Mansions

A regular Mansion with a front yard. This could be explored in the E3 Demo.
A similar Mansion with a somewhat different layout. It rains outside.
    A Mansion covered in snow.

Items and Treasures

    Coins
    Bills
    Treasure Chests
    Keys
    Gold Bars

Enemies

(Some Ghosts have sunglasses which protect them from the strobe light. The sunglasses must be vacuumed before capture.)

Unnamed green ghosts with 10 HP. (May appear with sunglasses)
Unnamed black ghost with 40 HP.
Unnamed red ghosts with 30 HP. (May appear with sunglasses)
Unnamed mummy-like ghosts which rise from the ground.
Suits of armor which attack with their swords if Luigi goes near them.
Spider that is purple when it crawls down and brown when it runs on the floor.
    Gold Spiders
Gates that are similar to the vines that block doors from the original Luigi's Mansion. Unlike the vines, though, the gates not just block doors but also paths like the middle of a stairway.

Bosses

    Telekinetic Ghost with 75 HP.
    King Boo

Rooms

    Guard Hall
    Front Yard
    Study
    Lab
    Library
    Dining Room
    Foyer
    Common Hall
    Entrance
    Garage
    Secret Pocket
    A pyramid themed room.
A Courtyard of some sort. It features Plants which give a key if they are flashed on with the strobe light.
    A Greenhouse
    A Bathroom
    A Lab like room
A Room with blue floating orbs with the door blocked by glass
A snow filled room similar to the Cold storage Room in the First game
Another bathroom where Luigi can sit on the toilet and the wall turns backward into another Room
A living room where Luigi gets sucked into the chimney
A bedroom with giant spider-webs Luigi can sit on the bed and it mysteriously flips over
A cellar type area with sand piles that slow Luigi down similar to the cellar in the first game

The Toad may just be a checkpoint like the last game.
I think that Toad wants to help out Luigi to me.

Can't all of these things happen at one mansion, or did Nintendo confirm multiple mansions?

How is saving not helpful?
Nintendo has confirmed at least 3 mansions as said earlier. I think you can save your game from the Pause Menu, just I can see Toad in the picture following Luigi.

Okay, see now I get what you mean, but then the question comes to, what exactly does/will/would Toad do?

His role is not yet known, we might have to wait for the next update from E3 or something else...

I'm really hoping we'll get some info  on this game this E3.  It's disappointing how Mario Tennis Open was just announced a couple of months ago and is now released while this game was announced last year and we still have no info on it.

It's likely that since Mario Tennis Open is a spin-off game, it's easier to make; Luigi's Mansion 2 was likely in early development (the demo must've been the first few levels, and even those are likely to be revised further) and still being developed right now.
